The folk-comedy duo, whose show ran on the HBO cable channel network for two seasons, will headline the festival's Friday night line-up, with Father John Misty, Ray Lamontagne and Raury also on the bill.
Their last performance together was at the Funny or Die Oddball Festival in 2013. They also performed for three nights at Auckland's Vector Arena in 2012.
Last year, Clement told The Guardian a Flight of the Conchords movie was "definitely a couple of years away, at least".
"The idea that Bret and I have been talking about ... would be a musical that doesn't have that old-fashioned, 'musical' feel," Clement said.
McKenzie has gone onto fame writing music for The Muppets movies, for which he won an Academy Award.
While Clement has starred in various films including People Places Things, Men in Black 3, and the upcoming the Fleshlumpeater in The BFG.
